Tabletop Exercise on Collaborative Approach to Zika Outbreak Response

Overview
Global interconnectedness between countries has contributed to the increased transmission of global emerging and reemerging infectious diseases. International travel across borders has become the main risk factor for disease spread from one place to another. Each person returning from a country with active local transmission is a risk factor for the entire country he/she is visiting after. One of the current infectious diseases threat that has travelled across continents is Zika virus. Data shows that prior 2015, Zika virus outbreaks have occurred in some areas of Africa, South East Asia and the Pacific Islands. The Zika virus outbreak is an example to call for a coordinated approach across borders and geographical regions in order to manage risk and mitigate against large outbreaks, as has been seen in the Americas. In responding to this need, Tabletop Exercise on Collaborative Approach to Zika Outbreak Response is organized to equip the academia with skills to develop Zika Outbreak Response Plan that increase South East Asia Capacity to deal with a pandemic, and to identify key persons from various multi-sectoral agencies in the region.

Trainer:
1. Prof. Wiku Adisasmito, DVM, M.Sc, PhD
A Professor at Faculty of Public Health, Universitas Indonesia. The coordinator of INDOHUN and Expert Panel of National Commission for Zoonotic Control.
2. Prof. Amin Soebandrio, MD, PhD, Sp.MK(K)
A Professor at Faculty of Medicine, Universitas Indonesia. The chairman of Eijkman Institute and Expert Panel of National Commission for Zoonotic Control.

This Training will provide participants with technical skills to:
1. Identify the policy issues associated with a Zika emergency.
2. Identify gaps in South East Asia countries preparedness.
3. Recognize the roles of government officials within a country (case study in Indonesia). Evaluate, and assess capacity needs for operational aspects of the regional and national response to Zika outbreak
4. Develop a respond plan to tackle ZIKA emergency in Indonesia as a case study.

The training is taught through a combination of lectures and exercise that prepare the participants with clear understanding of stakeholders’ roles and responsibilities that involve in responding the outbreak.

Ideal Participants
The targeted participants are Academia, Bachelor or Master or Doctoral students, health practitioners, policy maker, stakeholder with background from public health, medicine, and veterinary medicine in Indonesia, Thailand, Vietnam, and Malaysia

System Assessment for Disease Outbreak Preparedness Training

Overview
The world has experienced challenging health issues, particularly in emerging and re-emerging infectious diseases that bring potential harm to human being, animal, and ecosystem. Threats from infectious diseases need to be addressed at the human-animal-environmental interface and require strong health system that can support disease outbreak preparedness. South East Asia is a zoonosis hotspot and enhanced reporting and timely investigation of all suspected notifiable infectious disease threats within the region is vital. One of the current infectious diseases threat that threaten the region is Rabies. The increased Rabies cases is an example to call for a coordinated approach across borders and geographical regions in order to early detect the potential outbreak and develop disease preparedness plan. In responding to this need, System Assessment for Disease Outbreak Preparedness workshop is organized to equip the capability of stakeholders in collaboration with academia and health practitioners to collaborate multi-sectoral and multi-discipline and develop systems-based maps of agency, as well as develop operating procedures for One Health work within their network.

  4. Develop a disease outbreak preparedness plan

The training is taught through a combination of lectures and exercise that prepare the participants with clear understanding of stakeholders’ roles and responsibilities that involve in disease outbreak preparedness

Ideal Participants
The targeted participants are Stakeholder, Health Practitioners, Academia, Bachelor or Master or Doctoral students with background from or work in field of public health, medicine, and veterinary medicine in Indonesia, Thailand, Malaysia, and Vietnam.

Workshop of Drug Modeling

Drug modellingVirtual screening methods are well established in the modern drug discovery process since they significantly impact the efficient discovery of novel bioactive compounds. In addition, virtual screening with 3D-chemical feature-based pharmacophore models has been established as astate-of-the-art technique not only for elucidating key interactions between small molecules and proteins but also to identify active compounds with diverse scaffolds from a database of compounds.

LigandScout, a pharmacophore modeling and virtual screening platform, has been developed at Inte:Ligand to rapidly and efficiently interpret protein-ligand interactions and subsequently transform this information into 3D-chemical feature based pharmacophore models. In addition, these models can be generated in the absence of active-site data using a set of known active ligands to generate a rational mode of binding shared by the ligand set. Using these models in virtual screening campaigns are essential for identifying other compounds that can make the same interactions in an active site as well as understanding structure-activity relationships by transparently deriving key features for target interaction.

PyRx is a Virtual Screening software for Computational Drug Discovery that can be used to screen libraries of compounds against potential drug targets. PyRx enables Medicinal Chemists to run Virtual Screening from any platform and helps users in every step of this process – from data preparation to job submission and analysis of the results. While it is true that there is no magic button in the drug discovery process, PyRx includes docking wizard with an easy-to-use user interface which makes it a valuable tool for Computer-Aided Drug Design. PyRx also includes chemical spreadsheet-like functionality and powerful visualization engine that are essential for structure-based drug design.

In this workshop we will derive both structure- and ligand-based pharmacophores using PyRx and LigandScout and use them for fast and accurate virtual screening. We will practice model validation and predictivity assessment using Receiver Operating Characteristic (ROC) curves and enrichment factor (EF) analyses. Participants will be challenged to create and edit their own models with the aim of improving ROC curves and enrichment factors. Resulting models and metrics will be discussed in class to assess their suitability for virtual screening campaigns of untested compounds.

After attending the workshop, participants are expected to be able to:

  • Describe the basic of drug development methods: Stucture Base Drug Design (SBDD), Ligand Based Drug Design (LBDD), Fragment Based Drug Desig (FBDD)
  • Identify the virtual screening problem and choose the appropriate methods
  • Validate the virtual screening method through case study
  • Analyse and interpret the virtual screening result through case study
